Introduction
The HiPure Universal DNA Kit (D3018) is designed for purification of genomic DNA from a wide range of biological samples using silica membrane spin column technology. The optimized extraction chemistry supports efficient DNA recovery from complex sample matrices including blood, tissue, buffy coat, body fluids, swabs, dried blood spots and sperm samples.
The kit combines Proteinase K digestion with silica membrane adsorption to efficiently release genomic DNA from biological samples while removing proteins, lipids and other contaminants. The resulting DNA is suitable for routine molecular biology workflows such as PCR amplification, genotyping analysis and HLA typing studies.
Within the Magen DNA extraction portfolio, the HiPure Universal DNA Kit is positioned as a practical column-based solution for laboratories handling mixed sample types in routine workflows. Rather than being optimized for a single matrix, it provides a unified purification route across multiple common sample types, which is useful when workflow simplicity and broad sample compatibility are both required.

Workflow

Workflow Overview
The HiPure Universal DNA Kit uses a silica column-based workflow for purification of total DNA from routine sample types including tissue, blood, body fluids, cultured cells and swab-based materials. After sample-specific pretreatment, DNA is released by protease-assisted lysis, captured on the silica membrane under ethanol-containing binding conditions, and recovered through washing, drying and elution.
Sample Handling Logic
The main variation between sample types lies in the pretreatment and digestion stage. Tissue samples require cutting and complete digestion, cultured cells require pellet resuspension before lysis, while blood and other liquid samples can usually enter lysis more directly. Once the binding conditions are established, the downstream purification steps remain largely consistent.
Time and Workflow Characteristics
Under typical manual operation, the overall workflow for routine samples is usually completed within about 30–90 minutes, depending mainly on sample pretreatment and digestion requirements. For detailed sample-specific conditions, workflow guidance and estimated processing times, please refer to the Sample-Specific Workflow Note in the Download section.

Engineering Characteristics
Broad sample compatibility
The extraction chemistry supports purification of genomic DNA from multiple biological sample types including whole blood, buffy coat, tissue homogenates, body fluids, swabs and dried blood spot samples. This flexibility makes the system suitable for laboratories handling diverse sample sources.
Proteinase K digestion workflow
Biological samples are lysed using Proteinase K and optimized lysis buffers to efficiently release genomic DNA from cellular structures and chromatin complexes.
Silica membrane DNA adsorption
Genomic DNA selectively binds to the silica membrane under optimized binding conditions while proteins and cellular contaminants are removed during washing steps.
Stable purification performance
The column membrane and buffer chemistry are optimized to maintain consistent DNA recovery across different biological sample matrices.
Standard laboratory centrifuge operation
The spin-column workflow allows extraction using routine laboratory centrifuges without requiring specialized purification instruments.
Technical Validation
Validation experiments using multiple biological sample types demonstrated stable genomic DNA purification with purity ratios typically A260/280 around 1.7–1.9, indicating high-quality genomic DNA suitable for downstream molecular analysis workflows.
Extraction experiments across different tissue samples showed reproducible DNA yields with variation between replicate extractions typically below 10%, demonstrating stable purification performance across multiple sample types.
Purified genomic DNA produced clear high-molecular-weight bands in agarose gel electrophoresis and supported successful PCR amplification of target gene fragments, confirming compatibility with common molecular biology applications such as PCR amplification and genotyping analysis.
Comparative testing against commonly used commercial genomic DNA purification systems showed comparable DNA purity and recovery performance, supporting reliable DNA preparation for routine laboratory workflows.
Typical Applications

Storage and Stability
Proteinase K should be stored at 2-8°C upon arrival. However, short-term storage (up to 12 weeks) at room temperature (15-25°C) does not affect their performance. The remaining kit components can be stored at room temperature (15-25°C) and are stable for at least 18 months under these conditions. The entire kit can be stored at 2-8°C, but in this case buffers should be redissolved before use. Make sure that all buffers are at room temperature when used.
