How to read this note

1. Workflow structure
This workflow separates sample pretreatment from the shared downstream purification steps. It is intended as a practical companion to the product manual rather than a replacement for the official protocol. The downstream steps follow a magnetic bead-based purification workflow. This page uses a representative 5 mL plasma workflow, while the kit itself supports 1–8 mL plasma or serum input. For 1–4 mL inputs, the same workflow logic applies, but binding can usually be completed in a single loading step. For sample volumes above 4 mL, binding is performed in two sequential steps.

2. Time interpretation
Protocol times stated in the product manual are retained where applicable. Steps without explicit timing are estimated for an experienced operator. This workflow includes a 30–60 min incubation step for lysis, with additional handling time before and after incubation, followed by multi-step magnetic binding, washing, drying and elution procedures.

3. Workflow characteristics
This workflow is designed for large-volume cfDNA extraction using high-binding magnetic particles. Compared with column-based workflows, DNA capture is achieved through extended mixing with magnetic particles rather than membrane flow-through. Carrier RNA is optional but recommended, especially for low-abundance cfDNA samples, to reduce adsorption to consumables and stabilize yield. For sample volumes above 4 mL, lysate handling is performed in two sequential binding steps to maintain efficient particle-based recovery.

4. Practical considerations
For lower-volume magnetic cfDNA workflows (200–600 μL), refer to MagPure Circulating DNA Mini Kit (Cat. No. IVD5432).
