These cutoff values are described in the protocol as performance references based on marker-fragment testing. Users can adjust the Selection Solution volume according to their own validation results.

How to read this note

1. Workflow structure
This workflow separates sample pretreatment from the shared downstream purification steps. It is intended as a practical companion to the product manual rather than a replacement for the official protocol. The downstream steps follow a magnetic bead-based enrichment workflow rather than a standard total-cfDNA extraction workflow. This page uses a representative 5 mL plasma workflow.

2. Time interpretation
Protocol times stated in the product manual are retained where applicable. Steps without explicit timing are estimated for an experienced operator. This workflow includes one lysis stage, one large-fragment removal stage and one short-fragment capture stage, followed by washing, drying and two-step elution.

3. Workflow characteristics
This system is designed for selective enrichment of shorter cfDNA fragments through size-dependent magnetic bead separation. MagBind Particles are first used to capture larger DNA fragments, which are discarded. The remaining supernatant is then processed with MagPure Particles G to enrich shorter circulating DNA fractions.

4. Practical considerations
For standard vacuum-based cfDNA column workflows, refer to HiPure Circulating DNA Kit (Cat. No. IVD3182).
