Plasmid DNA Extraction Systems

Plasmid DNA purification should be selected according to plasmid copy number, culture volume, required yield, endotoxin level and processing format. Magen provides a structured plasmid DNA extraction system covering routine mini-prep, high-concentration preparation and large-scale plasmid production. The portfolio includes spin-column, filter-assisted and magnetic bead workflows for manual centrifugation, vacuum processing, 96-well handling and automated extraction platforms.

The system covers three practical purification levels. Standard molecular biology-grade plasmid DNA supports cloning, PCR, restriction digestion and routine sequencing. Low-endotoxin plasmid DNA below 1 EU/µg is intended for cell transfection and other endotoxin-sensitive in vitro workflows. Selected column products include an additional endotoxin-removal process to obtain endotoxin-free plasmid DNA below 0.1 EU/µg for animal injection.

Different purification chemistries are used to match plasmid behaviour rather than applying one binding system to every vector. Alcohol-mediated workflows provide broad plasmid recovery and high output for routine and high-copy vectors. High-salt-mediated binding improves RNA control and produces more representative OD-based concentration measurements, making it particularly suitable for medium- to low-copy plasmids. For high-copy vectors requiring very high total yield or concentrated recovery, cation-mediated selective plasmid precipitation is combined with column or magnetic separation so that output does not depend solely on the fixed adsorption capacity of a conventional silica membrane.

Within the portfolio, HiPure Plasmid Mini Kit (P1001) and MagPure Plasmid Mini Kit (P1811) serve as the standard column and magnetic reference products. HiPure Plasmid EF Maxi Kit (P1156) is the core routine low-endotoxin column system, while MagPure Plasmid EF Mini Kit (P1814) provides the corresponding magnetic route. HiPure Plasmid EF Midi Plus Kit (P1231) is positioned for high-concentration preparation, and HiPure HY Plasmid EF Mega Kit (P1116) serves as the core high-yield production route. Supporting products extend each branch by culture scale, throughput and instrument configuration.
