Introduction
The MagPure Fast Blood DNA Kit provides a rapid magnetic bead–based workflow for genomic DNA purification from whole blood and related biological fluids. Magnetic particles bind DNA released during sample lysis, allowing contaminants to be removed through simple magnetic separation steps.
The protocol is optimized for automated nucleic acid extraction instruments and supports processing of approximately 200–300 μl whole blood. The workflow is particularly suitable for laboratories operating medium-throughput extraction systems such as 16- or 32- channel magnetic extractors.
Within the Magen blood DNA extraction portfolio, laboratories performing routine column purification may use the HiPure Blood DNA Mini Kit (D3111), while universal automated workflows are supported by the MagPure Universal DNA Kit (IVD3102).

Technical Validation
The MagPure Fast Blood DNA Kit was validated using 200 µL whole blood samples on a MagMix 32 automated extraction system. The extracted DNA showed A260/280 values mainly around 1.78–1.90 and A260/230 values mainly around 1.7–2.0, indicating reliable DNA purity for automated blood DNA extraction workflows.
Workflow robustness was evaluated under different heating conditions. Both heated and non-heated sample-well conditions produced recoverable DNA with clear gel bands, supporting flexible use in automated extraction programs. Compared with the reference D6315 workflow, D6310 showed good recovery from samples containing partially degraded DNA.
The effect of lysis buffer volume was also tested using 200 µL blood input. Different Buffer MLA volumes produced usable DNA, and the validation suggested that 1.5× lysis buffer volume supported strong A260/230 performance. In the D6310 / D6310 prefilled format, the use of 500 µL Buffer MLA in the sample well supports flexible blood input volumes from approximately 100–400 µL.
Proteinase K input was further evaluated from 0–20 µL. The extraction workflow was not highly sensitive to Proteinase K amount, and acceptable DNA could still be recovered without Proteinase K under the tested conditions. However, adding Proteinase K improved yield and purity, with 5–10 µL providing high-quality DNA in the tested workflow.

Storage and Stability
Proteinase K Powder should be stored at 2–8°C upon arrival. However, short-term storage (up to 12 weeks) at room temperature (15–25°C) does not affect their performance. The remaining kit components can be stored at room temperature (15–25°C) and are stable for at least 18 months under these condition.
