Introduction

The HiPure Tissue & Blood DNA Midi Kit provides a higher-capacity column purification workflow for genomic DNA isolation from tissue samples, cultured cells, and whole blood. The silica membrane adsorption chemistry enables efficient binding of DNA released during protease digestion while proteins and contaminants are removed through sequential wash steps.

Compared with mini spin-column workflows, the Midi column format allows processing of larger biological samples such as up to 2 ml whole blood or approximately 200 mg tissue in a single purification procedure. The resulting DNA is suitable for PCR, Southern blotting, and other downstream molecular analysis.

For routine small-volume extraction workflows laboratories may refer to the HiPure Universal DNA Kit (D3018) or HiPure Blood DNA Mini Kit (D3111), while automated laboratories may use the MagPure Universal DNA Kit (IVD3102).

Principles

This product is based on silica column purification. The sample is lysed and digested with lysate and protease, DNA is released into the lysate. Transfer to an adsorption column. Nucleic acid is adsorbed on the membrane, while protein is not adsorbed and is removed with filtration. After washing proteins and other impurities, Nucleic acid was finally eluted with low-salt buffer (10mm Tris, pH9.0, 0.5mm EDTA).

Storage and Stability

Proteinase K, RNase A should be stored at 2-8°C upon arrival. However, short-term storage (up to 12 weeks) at room temperature (15-25°C) does not affect their performance. The remaining kit components can be stored at room temperature (15-25°C) and are stable for at least 18 months under these conditions.
