Introduction

The HiPure Tissue & Blood DNA Maxi Kit extends the column-based genomic DNA purification workflow to large sample volumes. The Maxi column format allows processing of substantially larger biological materials while maintaining the same silica membrane adsorption chemistry used in standard HiPure DNA purification systems.

The kit supports purification of genomic DNA from up to 10 ml whole blood or approximately 1 g tissue, enabling preparation of high DNA yields required for large-scale genomic studies, reference DNA preparation, or archival DNA storage.

Within the Magen DNA extraction systems this kit represents a large-volume column workflow. Laboratories performing routine DNA isolation may use the HiPure Universal DNA Kit (D3018), while automated high-throughput laboratories may adopt the MagPure Universal DNA Kit (IVD3102).

Principle

This product is based on silica column purification. The sample is lysed and digested with lysate and protease, DNA is released into the lysate. Transfer to an adsorption column. Nucleic acid is adsorbed on the membrane, while protein is not adsorbed and is removed with filtration. After washing proteins and other impurities, Nucleic acid was finally eluted with low-salt buffer (10mm Tris, pH9.0, 0.5mm EDTA).

Storage and Stability

Proteinase K, RNase A should be stored at 2-8°C upon arrival. However, short-term storage (up to 12 weeks) at room temperature (15-25°C) does not affect their performance. The remaining kit components can be stored at room temperature (15-25°C) and are stable for at least 18 months under these conditions.
