Tissue/Cell RNA Kits Selection Guide

Tissue/Cell RNA Extraction Overview

RNA extraction from tissues and cultured cells requires different strategies depending on sample composition, RNA type and downstream application. Soft tissues and cultured cells can often be processed using standard column-based workflows, while fibrous, lipid-rich or otherwise difficult tissues may require stronger lysis chemistry. In addition, some applications require only large RNA, while others require recovery of microRNA or more complete removal of genomic DNA.

Magen provides a structured tissue and cell RNA extraction portfolio covering silica membrane, magnetic bead and MagZol-based workflows. These systems support routine total RNA purification, microRNA isolation, high-throughput processing and automated extraction from a wide range of biological samples.

This guide is designed to help laboratories and distributors select the most appropriate RNA extraction workflow based on sample type, RNA target and downstream application requirements.

RNA Handling Considerations

RNA extraction results depend not only on the selection of an appropriate extraction system, but also on proper sample handling and RNase control throughout the workflow.

RNA is highly susceptible to degradation by RNases, which are widely present in the environment, laboratory surfaces and reagents. To ensure reliable results, it is critical to use RNase-free consumables, maintain clean working conditions and minimize sample handling time during extraction.

For low-abundance RNA such as circulating RNA or microRNA, careful handling becomes even more important, as degradation or contamination can significantly affect downstream analysis.

Quick Selection Guide

l If you need routine total RNA extraction

- Routine total RNA extraction from soft tissue or cultured cells → HiPure Total RNA Kit (R4011)

- Routine RNA extraction with reduced gDNA carryover → HiPure Total RNA Plus Kit (R4111)

- DNA-sensitive RNA applications → HiPure Total RNA Kit (IVD4121)

- High-throughput column-based RNA extraction → HiPure Total RNA 96 Kit (R4016)

l If your samples are difficult or heterogeneous

- Fibrous, hard-to-lyse or matrix-rich samples requiring MagZol front-end lysis → HiPure Universal RNA Kit (R4130)

- Difficult universal samples requiring magnetic handling after MagZol cleanup → MagPure Universal RNA Kit II (R6623)

- Protein-rich or viscous lysates where direct magnetic workflow is preferred → MagPure Total RNA Kit (R6622)

l If you need miRNA / small RNA

- miRNA-inclusive or miRNA-enriched RNA recovery from cells / tissues without phase separation → HiPure Cell miRNA Kit (R4311)

- Total RNA including miRNA, or separate large RNA and small RNA fractions requiring MagZol front-end lysis → HiPure Universal miRNA Kit (R4310)

- DNA-sensitive total RNA route including miRNA → HiPure Total RNA Kit (IVD4121)

l If you need automation

- Standard automated magnetic RNA purification with DNase support → MagPure Universal RNA Kit (IVD3020)

- Continuous automated workflow without mid-run pause → MagPure Universal RNA Kit B (IVD3020B)

- Magnetic workflow with Proteinase K digestion support → MagPure Total RNA Kit (R6622)

- MagZol front-end cleanup plus magnetic purification → MagPure Universal RNA Kit II (R6623)

Note: Routine total RNA workflows are generally larger-RNA-oriented unless the protocol is specifically designed for miRNA-inclusive recovery or small-RNA fractionation. For miRNA or small RNA studies, use a workflow validated for small RNA retention or enrichment.

Technical Note on A260/230 Interpretation

In low-input or low-yield RNA extraction workflows, A260/230 values may appear lower than expected and should not be used as the sole criterion for evaluating extraction performance. For column-based RNA purification, background absorbance from chaotropic buffer components can have a greater impact when RNA concentration is low.

By contrast, apparently higher A260/230 values do not always indicate better RNA quality. RNA quality should be interpreted together with yield, integrity and downstream assay performance such as reverse transcription, qRT-PCR or sequencing compatibility.

For this reason, Magen recommends evaluating RNA samples based on overall downstream suitability rather than relying on A260/230 alone, especially for low-abundance RNA samples.
