In addition, some applications require only large RNA, while others require recovery of microRNA or more complete removal of genomic DNA.

RNA extraction results depend not only on the selection of an appropriate extraction system, but also on proper sample handling and RNase control throughout the workflow.
RNA is highly susceptible to degradation by RNases, which are widely present in the environment, laboratory surfaces and reagents. To ensure reliable results, it is critical to use RNase-free consumables, maintain clean working conditions and minimize sample handling time during extraction.
For low-abundance RNA such as circulating RNA or microRNA, careful handling becomes even more important, as degradation or contamination can significantly affect downstream analysis.

Note: Standard total RNA kits typically recover RNA fragments larger than approximately 200 nt. For microRNA or other small RNA applications, dedicated small RNA kits should be selected.
In low-input or low-yield RNA extraction workflows, A260/230 values may appear lower than expected and should not be used as the sole criterion for evaluating extraction performance. For column-based RNA purification, background absorbance from chaotropic buffer components can have a greater impact when RNA concentration is low.
By contrast, apparently higher A260/230 values do not always indicate better RNA quality. RNA quality should be interpreted together with yield, integrity and downstream assay performance such as reverse transcription, qRT-PCR or sequencing compatibility.
For this reason, Magen recommends evaluating RNA samples based on overall downstream suitability rather than relying on A260/230 alone, especially for low-abundance RNA samples.
