Introduction

The HiPure Tissue DNA Kit is designed for routine purification of genomic DNA from animal tissues and cultured cells using silica membrane spin column technology. The workflow supports rapid lysis and efficient removal of proteins and enzymatic inhibitors, allowing recovery of high-quality DNA suitable for PCR amplification, sequencing preparation, and genotyping studies.

The system is optimized for processing up to 25 mg tissue or approximately 5 × 10⁶ cultured cells per preparation, making it suitable for routine molecular biology laboratories working with standard tissue samples and cell culture models.

Within the Magen Blood & Tissue DNA extraction portfolio, this kit serves as a tissue-focused workflow. Laboratories processing diverse biological samples may alternatively use the HiPure Universal DNA Kit (D3018) or the automated MagPure Universal DNA Kit (IVD3102).

Principle

This product is based on silica Column purification. The sample is lysed and digested with lysate and protease, DNA is released into the lysate. Transfer to an adsorption column. Nucleic acid is adsorbed on the membrane, while proteinis not adsorbed and is removed with filtration. After washing proteins andother impurities, Nucleic acid was finally eluted with low-salt buffer (10mmTris, pH9.0, 0.5mm EDTA).

Storage and Stability

RNase A and Proteinase K should be stored at 2-8°C upon arrival. However, short-term storage (up to 24 weeks) at room temperature (15-25°C) does not affect its performance. The remaining kit components can be stored dry at room temperature (15-25°C) and are stable for at least 18 months under these conditions. The entire kit can be stored at 2-8°C, but in this case buffers should be redissolved before use. Make sure that all buffers are at room temperature when used.
