Introduction
The HiPure HP Plant DNA Kit (D3187) is designed for genomic DNA extraction from plant tissues with high levels of polysaccharides, polyphenols and secondary metabolites, where standard SDS-based workflows often result in low yield or co-purification of inhibitors.
This kit combines CTAB-based lysis with organic extraction and silica column purification to enable stable DNA recovery from complex plant matrices. The workflow is particularly suited for woody plants, mature tissues and starch-containing samples that require additional impurity removal capacity.
Within the Magen plant DNA system, D3187 serves as the primary solution for complex plant samples, while routine samples can be processed using D3164 and automated workflows are supported by D6351.

Extraction Principle
Plant tissues are first lysed using a CTAB-based buffer system designed to disrupt cell walls and solubilize nucleic acids while preventing co-precipitation of polysaccharides.
Polyphenols and secondary metabolites are removed through organic extraction and selective phase separation, allowing DNA to remain in the aqueous phase.
Under high-salt conditions, genomic DNA binds to the silica membrane, while residual proteins, pigments and inhibitors are removed through sequential washing steps. Purified DNA is then eluted for downstream applications.

Technical Validation
The HiPure HP Plant DNA Kit was validated using diverse plant samples, including woody plants rich in polyphenols and plant materials rich in polysaccharides. Genomic DNA extracted from willow, orange, peach, juniper, longan and pomegranate showed clear high-molecular-weight bands above 23 kb by agarose gel electrophoresis, indicating good DNA integrity from polyphenol-rich woody plant samples.
Additional testing was performed with polysaccharide-rich plant materials such as towel gourd leaves, aloe leaves, sugarcane leaves, banana leaves, sweet potato leaves, onion, potato and garlic. Extracted DNA showed A260/280 values mainly around 1.88–2.18, with yields ranging from approximately 5–32 µg under the tested conditions, supporting reliable purification from difficult plant matrices.
Extraction stability was evaluated using 16 fresh cotton leaf samples. The purified DNA showed consistent electrophoresis bands across the tested samples, with A260/280 values mainly within 1.75–1.87 and yields of approximately 12–17 µg, indicating stable recovery and purity across replicate plant tissue samples.
Overall, the validation data support the use of the HiPure HP Plant DNA Kit for high-purity genomic DNA extraction from crops, woody plants, polysaccharide-rich samples and other difficult plant materials, with DNA quality suitable for PCR, SSR, AFLP, RAPD, Southern blot and related molecular analysis workflows.

Storage and Stability
This product can be stored dry at room temperature (15-25°C) and are stable for at least 18 months under these conditions.
