Workflow Overview
The HiPure SF Plant DNA Kit uses an SDS-based silica column workflow for purification of total DNA from simple plant and fungal samples. After mechanical disruption by liquid-nitrogen grinding or bead-beating, the sample is lysed with Buffer SPL in the presence of RNase A. Buffer PS is then added to precipitate cell debris, proteins and polysaccharide-associated contaminants. After centrifugation, the clarified DNA-containing supernatant is adjusted with Buffer GW1 and loaded onto the silica membrane, where DNA is captured, washed, dried and eluted with low-salt buffer or water.
Sample Handling Logic
This workflow is designed for routine plant and fungal samples where a direct SDS-based lysis route is sufficient. The main sample-dependent variation occurs during disruption, lysis and clarification, especially for samples that are viscous, pigment-rich or difficult to disperse. Once the lysate has been clarified and the binding condition has been adjusted, the downstream column purification follows a consistent bind, wash, dry and elute workflow.
Time and Workflow Characteristics
Under typical manual operation, the workflow is usually completed within about 55–65 minutes, depending mainly on sample disruption, lysate viscosity and the need for repeated column loading. This route is suitable for laboratories that require a fast and straightforward plant DNA column workflow without organic extraction. Storage and Stability
RNase A should be stored at 2-8°C upon arrival. However, short-term storage (up to 24 weeks) at room temperature (15-25°C) does not affect its performance. The remaining kit components can be stored dry at room temperature (15-25°C) and are stable for at least 18 months under these conditions.

HiPure SF Plant DNA Kit (D3164) is suitable for routine plant samples with low polysaccharide and polyphenol content.
For inhibitor-rich or difficult plant samples, CTAB-based systems such as HiPure HP Plant DNA Kit (D3187) may provide improved DNA purity.
For high-throughput or automated workflows, magnetic bead–based systems such as MagPure Plant DNA Kit (D6351) can be considered.
