This product is based on silica Column purification. The sample is lysed and digested with lysate and protease, DNA is released into the lysate. Transfer to an adsorption column. Nucleic acid is adsorbed on the membrane, while protein is not adsorbed and is removed with filtration. After washing proteins and other impurities, nucleic acid was finally eluted with low-salt buffer (10mmTris, pH9.0, 0.5mm EDTA).
The HiPure DNA Micro Kit was validated using low-input blood and trace biological samples to evaluate DNA recovery from limited sample amounts. In a 12-sample test using 10 µL anticoagulated whole blood, agarose gel electrophoresis showed consistent genomic DNA bands across replicates, supporting stable DNA extraction from small-volume blood samples.
Further testing was performed with 0.5 µL, 1 µL, 2 µL, 5 µL and 10 µL whole blood inputs. After extraction, 50% of the purified DNA was used as PCR template for β-actin amplification. Clear PCR products were observed across the tested input range, indicating that DNA recovered from very small blood volumes was suitable for PCR analysis.
The kit was also evaluated with representative trace and forensic-type samples, including buccal swab, cigarette butt, chewing gum, saliva, blood stain and hair roots. Extracted DNA was analyzed by gel electrophoresis and PCR amplification, showing that the purified DNA could be used directly for PCR detection under the tested conditions.
Carrier-assisted recovery was tested using 5 µL blood samples and diluted DNA marker fragments in serum. The results showed improved DNA recovery when Carrier Mix was included, supporting the use of carrier-assisted binding for very low-input samples where target DNA amount is limited.

Carrier RNA and Proteinase K should be stored at 2–8°C upon arrival. However, short-term storage (up to 12 weeks) at room temperature (15–25°C) does not affect its performance. The remaining kit components can be stored dry at room temperature (15–25°C) and are stable for at least 18 months under theseconditions.
