Introduction

HiPure Plasmid EF Midi Kit is positioned for mid-scale low-endotoxin preparation from medium- to high-copy plasmids, especially when concentrated plasmid recovery is needed without moving directly into a large-format maxi workflow.

Within the EF branch, P1231 sits between P1154 and P1156B. HiPure Plasmid EF Mini Kit(P1154) remains the compact EF mini option for lower-input workflows, while HiPure Plasmid EF Maxi Kit B (P1156B) is the low-endotoxin column reference model for larger preparation volumes. Laboratories working in the standard non-EF route at comparable or higher scale may instead refer to HiPure Fastfilter Plasmid Midi Kit(P1013) or P1014.

Principle

The workflow combines alkaline lysis, clarified lysate handling and selective precipitation before low-endotoxin column purification. The system uses a compact column-with-extender format that supports mid-scale plasmid preparation with relatively small elution volume, helping recover plasmid DNA at higher concentration than larger-format workflows designed mainly for total output.

Technical Validation

Internal validation showed A260/280 values around 1.8–1.9 and A260/230 values around 1.8–2.8. In medium- to high-copy plasmid workflows, total plasmid output increased across 10–100 mL culture input and reached up to 563 µg, supporting its role as a concentrated mid-scale low-endotoxin preparation format. Restriction digestion and short-term room-temperature stability testing further supported downstream usability.

In internal comparative testing with common midi-scale column workflows, P1231 showed comparable purity and practical plasmid recovery across several routine plasmid vectors.

Storage and Stability

The kit components can be stored dry at room temperature (15-25°C) and are stable for at least 18 months under these conditions. If any precipitates form in the buffers, warm at 37 to dissolve. After addition of RNase A and optional LyseBlue reagent, Buffer P1 is stable for 6 months when stored at 2–8°C.
