Workflow Overview
The HiPure Plant RNA Mini Kit uses a dual-column silica workflow for purification of high-quality RNA from simple plant samples. Plant tissue is first disrupted under liquid nitrogen and lysed with Buffer RLC or Buffer PRC1 in the presence of β-mercaptoethanol or DTT. The clarified lysate is passed through a gDNA removal column, where genomic DNA is reduced while the RNA-containing flow-through is collected. Ethanol is then added to the flow-through to establish RNA binding conditions, and the sample is loaded onto the RNA column for binding, washing, drying and elution.
Sample Handling Logic
This workflow is designed for routine plant RNA extraction where operational simplicity and upstream gDNA reduction are important. Buffer RLC is used for most simple plant samples, while Buffer PRC1 provides an alternative for samples that may be affected by secondary metabolites or lysate solidification. The main sample-dependent variation occurs during liquid-nitrogen grinding, lysate clarification and passage through the gDNA removal column. Once the RNA-containing flow-through has been adjusted with ethanol, the downstream column workflow remains consistent.
Time and Workflow Characteristics
Under typical manual operation, the workflow is usually completed within about 35–45 minutes, depending mainly on sample disruption, lysate clarity and loading volume. Optional on-column DNase digestion can be added when downstream applications require more stringent DNA control. This route is suitable for laboratories that need a straightforward dual-column plant RNA workflow with gDNA reduction before RNA purification. Storage and Stability
The Kit can be stored dry at room temperature (15-25°C) and are stable for at least 18 months under these conditions. Make sure that all buffers are at room temperature when used. During shipment, crystals or precipitation may form in the Buffer RLC/PRC1. Dissolve by warming buffer to 37°C.

HiPure Plant RNA Mini Kit (R4151) is suitable for routine plant samples with low polysaccharide and polyphenol content.
For unknown or variable samples, HiPure Plant RNA Plus Kit (R4150) is recommended as a more broadly compatible system.
For difficult plant samples, CTAB-based workflows such as HiPure HP Plant RNA Mini Kit (R4165) may provide improved performance.
