Introduction

The MagPure Blood RNA Kit (R6611) is designed for purification of total RNA from whole blood samples using magnetic bead purification technology. Blood samples present specific challenges for RNA extraction due to the presence of RNases and other contaminants that can degrade RNA molecules during sample processing.

The magnetic bead purification workflow enables efficient RNA recovery from anticoagulated blood, buffy coat and bone marrow samples. Magnetic separation allows automated washing and elution steps, supporting consistent RNA purification in automated extraction workflows.

Technical Validation

MagPure Blood RNA Kit was evaluated as a magnetic bead-based total RNA extraction workflow for anticoagulated blood samples. The workflow includes red blood cell lysis, leukocyte recovery, Buffer RTL lysis, magnetic particle RNA binding, DNase I digestion, magnetic washing and RNase-free water elution. The kit can be used in manual magnetic handling or adapted to automated magnetic extraction platforms such as KingFisher Flex.

Performance testing was performed using 14 fresh blood samples. RNA was extracted using the MagPure Blood RNA workflow, measured by Nanodrop 2000 and analyzed by 1.5% agarose gel electrophoresis. Across the tested samples, RNA concentrations ranged from 208.72–278.48 ng/µL, corresponding to total RNA yields of 20.87–27.85 µg under the tested elution conditions. A260/280 values remained consistent at 2.10–2.14, while A260/230 values ranged from 1.26–2.00, reflecting sample-dependent variation in blood RNA extraction.

Agarose gel electrophoresis showed clear RNA band patterns across all 14 blood samples, with no obvious degradation observed under the tested conditions. The inclusion of DNase I digestion in the workflow supports reduction of genomic DNA background before elution, which is important for downstream RNA-based assays that are sensitive to residual DNA.

Together, these results support R6611 as a magnetic bead-based blood RNA extraction workflow for laboratories requiring reproducible RNA recovery from anticoagulated blood, leukocyte-rich blood fractions or related hematological samples. The recovered RNA is suitable for downstream RNA analysis workflows such as RT-PCR, real-time PCR and sequencing-oriented library preparation, with final performance depending on sample condition, leukocyte content and downstream assay requirements.

Storage and Stability

DNase I should be shipped with ice pack or dry ice and stored at -20°C upon arrival. MagPure Particles N and Proteinase K should be stored at 2–8°C upon arrival. However, short-term storage(up to 8 weeks) at room temperature (15–25°C) does not affect their performance.The remaining kit components can be store at room temperature and are stable for up to 18 months under these conditions.
