The Kit combines the speed and efficiency of silica-based technology with the convenient handling of magnetic particles for purification of total RNA. Samples are lysed and RNA is purified from lysates in one step through its binding to the silica surface of the particles in the presence of a chaotropic salt. The particles are separated from the lysates using a magnet and DNA is removed by treatment with RNase-free DNase. The magnetic particles are efficiently washed, and RNA is eluted in RNase-free water.
MagPure Stool RNA Kit was evaluated as a magnetic bead-based RNA extraction workflow for stool and microbial samples. The workflow combines bead-tube homogenization, SPL / PHC lysis, chloroform-assisted clarification, magnetic particle RNA binding, DNase I treatment, magnetic washing and RNase-free water elution. The kit is designed for manual magnetic handling and can also be adapted to automated magnetic extraction platforms such as KingFisher Flex.
In stool sample testing, 300 mg stool input was homogenized in a bead tube with lysis reagents and phenol / chloroform / isoamyl alcohol, followed by centrifugation. A 300 µL clarified supernatant was processed using the R6626 magnetic workflow, and RNA was eluted in 70 µL. Across six replicate stool extractions, RNA yields were 74.24–82.04 µg, with A260/280 values of 2.14–2.17 and A260/230 values of 1.80–2.00 under the tested conditions. Agarose gel electrophoresis showed clear RNA band patterns without obvious degradation.
Bacterial RNA recovery was evaluated using concentrated Escherichia coli samples. A 300 µL bacterial suspension was processed through the same bead-tube lysis and R6626 magnetic purification workflow, with RNA eluted in 70 µL. Across six replicate bacterial extractions, RNA yields were 4.94–6.19 µg, with A260/280 values of 2.09–2.15 and A260/230 values of 1.71–1.96. Electrophoresis analysis showed clear RNA bands, supporting recovery of bacterial RNA using the magnetic bead-based workflow.
The R6626 workflow includes DNase I treatment during magnetic purification, supporting applications where residual genomic DNA background may interfere with RT-PCR or other RNA-based assays. Together, the stool and bacterial RNA data support R6626 as a magnetic, automation-ready stool RNA extraction workflow for laboratories that require scalable handling, inhibitor reduction and integrated DNA removal.

MagPure RNA Particles should be stored at 2–8°C upon arrival. DNase I should be stored at -20°C. However, short-term storage (DNase I up to 1 weeks, MagPure RNA Particles up to 8 weeks) at room temperature (15–25°C) does not affect their performance. The remaining kit components can be stored at room temperature (15–25°C) and are stable for at least 18 months under these conditions.
