Introduction
The MagPure Blood DNA Kit provides a magnetic bead–based workflow for purification of genomic DNA from anticoagulated whole blood samples. During extraction, DNA released from lysed blood cells is captured by magnetic particles and purified through automated washing steps, producing high-quality genomic DNA suitable for PCR amplification, genotyping analysis, and routine molecular testing.
The protocol is optimized for automated nucleic acid extraction instruments and is particularly suited for 96-channel extraction platforms, allowing efficient processing of large sample batches in clinical and research laboratories.
Within the Magen blood DNA purification portfolio, laboratories operating medium-throughput extraction systems (16–48 channels) may use the MagPure Fast Blood DNA Kit (D6310C), while column-based manual extraction workflows are available through the HiPure Blood DNA Mini Kit (D3111).

Principle
This product is based on the purification method of high binding magnetic particles. The sample is lysed and digested under the action of lysate and Protease. DNA is released into the lysate. After adding magnetic particles and binding solution, DNA will be adsorbed on the surface of magnetic particles, and impurities such as proteins will be removed without adsorption. The adsorbed particles were washed with washing solution to remove proteins and impurities, washed with ethanol to remove salts, and finally DNA was eluted by Elution Buffer.
Technical Validation
The MagPure Blood DNA Kit was validated using human whole blood samples on automated magnetic bead extraction platforms. In a large-scale extraction test, 200 µL human blood samples were processed using the MagPure Blood DNA Kit and KingFisher Flex. Selected NanoDrop results showed DNA yields of approximately 10–58 µg, with A260/280 values mainly within 1.81–1.92 and A260/230 values mainly within 1.5–2.5, supporting reliable DNA purity for high-throughput blood DNA extraction workflows.
Compatibility with different anticoagulants was evaluated using 200 µL human blood samples collected with EDTA, citrate and heparin. Agarose gel electrophoresis showed intact genomic DNA bands across the tested anticoagulant types, indicating that the workflow can produce high-quality blood DNA from common anticoagulated blood samples.
Low-volume blood extraction was also tested using 2 µL and 5 µL human anticoagulated blood diluted to 200 µL before extraction. Gel electrophoresis showed recoverable genomic DNA from these low-input samples, supporting efficient DNA recovery when available blood volume is limited.
Downstream compatibility was confirmed by PCR amplification of a 400 bp DNA fragment using DNA extracted from buccal swabs, dried blood spots and anticoagulated blood samples. Clear PCR products were obtained, indicating that DNA purified with the MagPure Blood DNA Kit can be used directly for PCR-based analysis. Automated processing on KingFisher Flex can complete 96 samples in approximately 40 minutes.

Storage and Stability
Proteinase K, MagPure Particles should be stored at 2-8°C upon arrival. However, short-term storage (up to 24 weeks) at room temperature (15-25°C) does not affect their performance. The remaining kit components can be stored dry at room temperature (15-25°C) and are stable for at least 18 months under these conditions. The entire kit can be stored at 2-8°C, but in this case buffers should be redissolved before use. Make sure that all buffers are at room temperature when used.
