Introduction
The HiPure HP Plant DNA Kit (D3187) is designed for genomic DNA extraction from plant tissues with high levels of polysaccharides, polyphenols and secondary metabolites, where standard SDS-based workflows often result in low yield or co-purification of inhibitors.
This kit combines CTAB-based lysis with organic extraction and silica column purification to enable stable DNA recovery from complex plant matrices. The workflow is particularly suited for woody plants, mature tissues and starch-containing samples that require additional impurity removal capacity.
Within the Magen plant DNA system, D3187 serves as the primary solution for complex plant samples, while routine samples can be processed using D3164 and automated workflows are supported by D6351.

Extraction Principle
Plant tissues are first lysed using a CTAB-based buffer system designed to disrupt cell walls and solubilize nucleic acids while preventing co-precipitation of polysaccharides.
Polyphenols and secondary metabolites are removed through organic extraction and selective phase separation, allowing DNA to remain in the aqueous phase.
Under high-salt conditions, genomic DNA binds to the silica membrane, while residual proteins, pigments and inhibitors are removed through sequential washing steps. Purified DNA is then eluted for downstream applications.
