Introduction

MagPure Bacterial HW DNA Kit is designed for high molecular weight DNA extraction from bacterial samples, where efficient disruption of the cell wall is a critical factor influencing DNA yield and integrity. Gram-positive and Gram-negative bacteria differ significantly in cell wall structure, and standard lysis conditions are often insufficient for consistent DNA recovery across different strains.

This kit incorporates enzymatic lysis strategies, including lysozyme-assisted digestion, to improve cell wall disruption while preserving genomic DNA integrity. The subsequent magnetic bead purification process is performed under controlled conditions to minimize shearing, enabling recovery of long DNA fragments suitable for downstream applications requiring high integrity.

For laboratories working with mixed or non-microbial samples, the MagPure Universal HW DNA Kit (D6381) provides a more general solution without specialized lysis steps. Tissue samples can be processed using MagPure Tissue HW DNA Kit (D6382), while fungal samples requiring mechanical disruption are better suited to MagPure Fungal HW DNA Kit (D6384). For plant-derived samples with complex secondary metabolites, MagPure Plant HW DNA Kit (D6385) provides dedicated inhibitor control strategies.

Storage and Stability

Lysozyme, Proteinase K, RNase A and MagPure Particles G2 should be stored at 2–8°C upon arrival. However, short-term storage (up to 24 weeks) at room temperature (15–25°C) does not affect their performance. The remaining kit components can be stored at room temperature (15–25°C) and are stable for at least 18 months under these conditions.
