Workflow Overview
The MagPure Pathogen DNA/RNA Kit B uses a low-background reagent system combined with magnetic bead–based purification for pathogen DNA/RNA extraction. After low-speed clarification when required, the sample is processed through SDS-assisted bead-tube disruption to support broader release of nucleic acids from mixed pathogen materials. The clarified lysate is then purified using MPN9 magnetic particles, followed by washing, drying and elution. This workflow is positioned for tNGS and pathogen panel–oriented applications where direct pathogen disruption and low reagent-derived nucleic acid background are important.
Sample Handling Logic
The workflow is suitable for pathogen-containing samples such as plasma, serum, body fluid, homogenate suspension, culture solution, cell suspension, soaking solution and concentrated pathogen solution. For cell-rich samples, low-speed clarification helps reduce excessive host-cell carryover while retaining liquid-phase pathogen material for direct disruption. For sputum or lavage samples, DTT liquefaction may be required before extraction. The workflow should be understood as a direct pathogen disruption route rather than a host-background reduction or pathogen enrichment workflow.
Time and Workflow Characteristics
Under typical manual operation by an experienced operator, the workflow is usually completed within about 50–65 minutes. Processing time depends mainly on sample conditioning, bead-tube disruption method and clarification requirements. Longer handling may be required when vortex-based disruption, DTT liquefaction or additional sample clarification is used. Storage and Stability
Particles MPN9 and Proteinase K should be stored at 2–8°C upon arrival. However, short-term storage (up to 8 weeks) at room temperature (15–25°C) does not affect their performance. The remaining kit components can be stored at room temperature (15–25°C) and are stable for at least 18 months under these conditions.
