Introduction
The MagPure Universal DNA Kit (IVD3102) is designed for automated purification of genomic DNA from diverse biological samples using magnetic bead purification technology. The extraction chemistry enables efficient DNA recovery from multiple sample types including whole blood, buffy coat, cultured cells, tissue homogenates and other biological materials commonly processed in molecular laboratories.
Magnetic bead purification allows selective binding of genomic DNA molecules while enabling automated washing and elution steps through magnetic separation. Compared with manual column workflows, magnetic bead extraction systems provide improved workflow consistency and support high-throughput nucleic acid purification.
The MagPure Universal DNA Kit is compatible with automated nucleic acid purification instruments and supports routine DNA extraction workflows in research laboratories, clinical molecular laboratories and high-throughput testing environments.

Magnetic bead-based DNA purification
The MagPure Universal DNA Kit uses high-binding magnetic particles to capture genomic DNA under optimized binding conditions. After sample lysis and proteinase digestion, DNA is bound to magnetic particles, while proteins, salts and other impurities are removed through sequential washing steps.
Universal sample processing design
The kit supports genomic DNA extraction from multiple biological sample types, including blood, saliva, swabs, blood spots, semen, tissue samples, cultured cells and FFPE-derived material. Sample-specific lysis conditions are used before the shared magnetic bead purification process, allowing different sample types to be processed within one extraction system.
Optimized lysis-to-binding workflow
Different biological samples require different pretreatment and lysis conditions before DNA binding. The workflow combines proteinase digestion, optimized lysis chemistry and magnetic bead adsorption to support stable DNA release and purification from routine and more challenging sample matrices.
Automation-ready format
The kit is designed for automated magnetic bead purification workflows and supports 96-well processing formats. This makes it suitable for laboratories that require standardized genomic DNA extraction across multiple sample types and higher-throughput processing needs.
Downstream-compatible genomic DNA
Purified genomic DNA is suitable for common molecular biology applications, including PCR, qPCR, genotyping, HLA typing and sequencing-related sample preparation.
Technical Validation
Validation experiments were performed to evaluate the DNA extraction performance of the MagPure Universal DNA Kit across representative biological sample types, including blood-derived samples, tissue samples, pig tail tissue and saliva samples. DNA quality was assessed by spectrophotometric measurement, fluorescence-based quantification and agarose gel electrophoresis where applicable.
Blood-derived sample validation
The kit was tested with blood-derived samples including whole blood, dry blood spots and coagulated human blood. For dry blood spot samples, DNA recovery was evaluated against a reference commercial extraction workflow using OD measurement, Qubit quantification and gel electrophoresis. The results showed reliable DNA recovery from low-input dried blood material and highlighted the importance of fluorescence-based quantification when OD readings may overestimate DNA concentration.
For coagulated or partially clotted blood samples, optimized sample liquefaction and magnetic bead purification improved extraction consistency. The validation data showed stable A260/280 values around the expected genomic DNA range and better A260/230 stability under the optimized workflow. Gel electrophoresis also showed clearer genomic DNA bands with less smearing when the coagulated sample was sufficiently dispersed before extraction.
Tissue sample validation
The kit was evaluated with multiple tissue types, including mouse ear, kidney, skin, tail, liver, spleen, heart and lung samples. Agarose gel electrophoresis showed successful genomic DNA recovery across the tested tissue matrices, supporting the use of the kit for routine tissue DNA extraction after appropriate lysis and proteinase digestion.
Pig tail tissue validation
A 96-sample pig tail tissue extraction test was performed using an automated magnetic bead workflow. Most samples produced measurable genomic DNA with A260/280 values within the expected range, supporting the kit’s use in routine animal tissue genotyping and high-throughput extraction workflows.
Saliva sample validation
Fresh and stored saliva samples were tested to evaluate performance with non-invasive biological samples. The 96-sample saliva extraction data showed consistent DNA recovery across most samples, with purity values suitable for routine downstream molecular analysis.
Overall, the experimental data support the use of the MagPure Universal DNA Kit as an automated magnetic bead-based genomic DNA extraction system for multiple validated sample types, including blood-derived samples, tissue, pig tail tissue and saliva.

Storage and Stability
Proteinase K, RNase A, MagPure Particles should be stored at 2-8°C upon arrival. However, short-term storage (up to 24 weeks) at room temperature (15-25°C) does not affect their performance. The remaining kit components can be stored at room temperature (15-25°C) and are stable for at least 18 months under these conditions.
