Introduction

MagPure Pathogen RNA Kit is designed for purification of pathogen RNA from sputum and similar respiratory samples. Microorganisms are first disrupted using bead-assisted mechanical agitation combined with chemical lysis buffers, releasing microbial RNA from bacterial cells.

DNase digestion is then applied to remove background DNA before RNA molecules are captured and purified using magnetic bead adsorption. The resulting RNA is suitable for downstream molecular analysis and microbial detection workflows.

This system is particularly useful for pathogen RNA detection in respiratory samples. For broader pathogen nucleic acid extraction workflows laboratories may use MagPure Pathogen DNA/RNA Kit (IVD6672), while sequencing-focused enrichment strategies are available with MagPure Pathogen DNA/RNA Enrich Kit (R6672C).

Principle

This product is based on the purification method of high binding magnetic particles. The sample is lysed and digested under the action of lysate and Protease. After adding magnetic particles and binding solution, DNA/RNA will be adsorbed on the surface of magnetic particles, and impurities such as proteins will be removed without adsorption. The adsorbed particles were washed with washing solution to remove proteins and impurities, washed with ethanol to remove salts, and finally DNA/RNA was eluted by Buffer NFW.

Storage and Stability

MagPure Particles and Proteinase K, DNase I  should be stored at 2–8°C upon arrival. However, short-term storage (up to 2 weeks) at room temperature (15–25°C) does not affect their performance. The remaining kit components can be stored at room temperature (15–25°C) and are stable for 18 months under these conditions.
